چکیده

Subsequent reports from our laboratory showed that both carnitine (/3-hydroxy-y-trimethylammonium butyrate) and acetylcarnitinei increased oxygen uptake and oxidation of long chain fatty acids by heart homogenates incubated under conditions optimal for fatty acid oxidation (2, 3). Acetylcarnitine action could not be attributed simply to formation of carnitine via hydrolysis because acetylcarnitine addition augmented respiration in the absence of added coenzyme A (CoA) whereas carnitine had no such effect (3). Further, acetylcarnitine enhanced the conversion of (Y-labeled fatty acids to COZ in the absence of CoA to a greater extent than carnitine did. The possibility that carnitine acetyltransferase and other transferases might be directly involved in mediating the metabolic effects of carnitine on fatty acid metabolism has been discussed in previous publications (3-5). In the present investigation we shall describe the properties of carnitine acetyltransferase and will give results demonstrating that the 0-acyl structure of acylcarnitine derivatives has a high group potential.

Deacylation of acetyl-coenzyme A and acetylcarnitine by liver preparations.

The breakdown of acetylcarnitine catalysed by extracts of rat and sheep liver was completely abolished by Sephadex G-25 gel filtration, whereas the hydrolysis of acetyl-CoA was unaffected. Acetyl-CoA and CoA acted catalytically in restoring the ability of Sephadex-treated extracts to break down acetylcarnitine, which was therefore not due to an acetylcarnitine hydrolase but to the sequential ac...

Carnitine acetyltransferase in nervous tissue.

Some of the kinetic properties of carnitine acetyltransferase in brain homogenates have been reported with the use of a new, sensitive radiometric method for the determination of acetylcarnitine. Those properties and the distribution pattern of carnitine acetyltransferase in various rat tissues, subcellular fractions, and discrete areas of the brain have been compared with those obtained for ch...

Structure of human carnitine acetyltransferase. Molecular basis for fatty acyl transfer.

Carnitine acyltransferases are a family of ubiquitous enzymes that play a pivotal role in cellular energy metabolism. We report here the x-ray structure of human carnitine acetyltransferase to a 1.6-A resolution. This structure reveals a monomeric protein of two equally sized alpha/beta domains. Each domain is shown to have a partially similar fold to other known but oligomeric enzymes that are...

Isolation and purification of mitochondrial carnitine octanoyltransferase activities from beef heart.

The purpose of this study was to characterize the mitochondrial carnitine octanoyltransferase of beef heart. Carnitine acyltransferase activities were solubilized from isolated beef heart mitochondria using KC1 and the nonionic detergent, Triton X-100, at final concentrations of 1 M and 2%, respectively. Upon fractionation of the solubilized protein on Cibacron Blue Sepharose, two protein peaks...

Choline acetyltransferase from rat brain.

The localization, purification, and some enzymatic properties of choline acetyltransferase from rat brain were studied. Most assays were performed with a specific radiometric micromethod. Solubilization of the enzyme was examined after homogenization of cerebral cortices by means which disintegrate nerve endings. In isotonic KC1 the enzyme was recovered in solution. In more dilute KC1 the enzym...
